摘 要:  采用体外细胞培养模式,通过测定细胞碱式磷酸酶(ALP)活性及细胞四甲基偶氮砜(MTT)还原能力, 对构成金属和合金生物材料的金属离子(Al, Ni, Cr, V, Ag)的毒性进行了定量评估,探讨了金属离子或原子对人体组织的毒害和腐蚀作用。结果表明微量六价铬离子表现出强烈的细胞毒性。当溶液离子度高于100μmol/L时,钒和镍亦表现强烈毒性。银离子和三价铬离子浓度高于500μmol/L后表现毒性。构成老年痴呆病成因的金属铝离子的毒性具有双向性。

Approach of metal cytotoxicity(Ⅰ)

Abstract:Cytoxicity analysis is the critical testing of evaluating biomaterials in the ASTM,ISO standards. The approach has invested in vitro the toxicity of metal ions (Al, Ni, Cr(Ⅲ), Cr(Ⅵ), V, and Ag) that construct the metallic biomaterials. By measurements of cell alkline activity (ALP) and reduction ability of cell MTT,  the toxicity of prevalence metallicbiomaterials has been studied,  and furthermore, the poison and erosion of metal ions and atoms on human tissue are discussed. The results display that trace Cr(Ⅵ) shows serious cytotoxicity, and V as well as Ni is cytotoxicity if the ion concentration in culture medium is over 100μmol/L. A strange phenomenon is that Ag is also cytotoxicity if the ion concentration is higher than 500μmol/L. Al ions is biphasic in cytotoxicity. At low ion concentration (<10μmol/L), Al ions can stimulate cell proliferation,  whereas concentration is over 1000μmol/L, cytotoxicity increases.
